【Animal Modeling】-How to establish a mouse model of intestinal flora derived from patients with coronary heart disease?

  Purpose: To establish a human mouse model of plant-associated fecal bacterial transplantation (HFA) in patients with coronary heart disease to evaluate the model.

  Methods: Twenty-eight infertile female C57BL/6J mice were randomly divided into control (CON) group and model (CAD) group. A group of 7 animals were divided into feces, blood, 16SDNA aorta, and blood lipids and cells. Factor, the examination was sent to pathology.

  Result: From the fifth week, the animal's body weight in the CAD group increased rapidly (P\u003c0.05). α diversity analysis, the Simpson index values (Pu003c0.05, Pu003c0.01), Shannon index (Pu003c0.05, Pu003c0.01), ACE (Pu003c0.01) u003c0.05 super 1 index at two time points in the CAD group Increase) is all decrease (P\u003c0.05). Reading the CON group, there are mainly Firmicutes, Proteobacteria, Bacteroides, Verrucomicrobia, Fusobacteria, Actinomycetes, Cyanobacteria, which have been distributed in Tenikut. Within six weeks of modeling, the abundance of soft wall bacteria in Firmicutes and CAD groups decreased (P\u003c0.05), and the abundance of Bacteroides and Verrucomicrobia increased (P\u003c0.01). Within 10 weeks of modeling, the abundance of Firmicutes has increased. In the CAD group (P\u003c0.01), the abundance of Bacteroides and Weruko microshuang has increased (P\u003c0.01). In the β diversity analysis, the types of the bowel CAD group and the CON group are distributed in different areas, and different stages of the same group have been shown to be distributed in the same area. There is a significant difference in the composition of the microbial species between the CAD group and the CON group (P\u003c0.05). In the CAD group, blood triglycerides (TG) (P\u003c0.05, P\u003c0.01), cholesterol (TC) (P\u003c0.05), lactate dehydrogenase (LDH) (P\u003c0. 0P\u003c0. 0001) and phosphokinase (CK) (P\u003c0.0P\u003c0.05) values increase both. Low-density lipoprotein (LDL-C) increased in six weeks modeled (P\u003c0.05). Cytokine detection, the increase in IL-6 concentration was modeled for 6 weeks (P\u003c0.05), and it was reduced by 10 weeks (P\u003c0.00001); IL-10 level was modeled to decrease within 10 weeks (P\u003c0. 05); IL-2, IL-4, IL-5, IL1β concentration increased within 10 weeks modeled (P\u003c0.000P\u003c0.05, P\u003c0.000P\u003c0 0.01). IL-12p70 and TNF-α had no significant difference in INF-γ at the two time points. CAD group and coronary artery hematoxylin CON group-In eosin (H.E) staining, atherosclerotic changes such as foam cell formation were not found in the two time points after modeling.

  Conclusion: In this study, using fecal bacterial transplantation, we acquired a mouse flora derived from patients with coronary heart disease. Intestinal flora imbalance, body weight, blood lipids, cytokine content and other indicators are rich in structure, showing similar clinic changes.
