Objective: To explore the factors affecting the establishment of a C57BL/6J mouse melanoma lung metastasis model, including the method of tumor inoculation, the number of cells inoculated and the cycle of tumor formation.
Methods: Culture mouse melanoma cells B16F10 in vitro; 1) Take 18 male mice aged 6-8 weeks and randomly divide them into three groups, each with 6 mice. The mice were injected via tail vein, intraperitoneal injection and subcutaneous injection. Mice were injected with 100 μL (3×106 cells) of B16F10 cell suspension. Two weeks later, the mice were dissected and the growth and metastasis of melanoma were observed; 2) Divide into 3 groups, the same as above, and inject 3×106 via the tail vein. Cells, 1×106 cells, 3×105 cells, two weeks later, the mice were dissected and the growth and metastasis of melanoma were observed; 3) Divide into 3 groups, same as above, inject 1×106 cells into the tail vein, respectively The mice were dissected in 1 week, 2 weeks, and 3 weeks to observe the growth and metastasis of melanoma.
Results: 1) When mouse melanoma cells were injected into the tail vein, the success rate of lung metastasis in mice was 100%, but no lung metastasis occurred in intraperitoneal injection and subcutaneous injection; 2) When the number of melanoma cells inoculated in mice was 1 × 106 , The number of melanoma cells that have metastasis to the lung is moderate; when the number of cells inoculated is 3×106, the number of melanoma cells that have metastasis to the lung is too much; when the number of cells is 3×105, the number of melanoma cells that have metastasis to the lungs The number is small; 3) 1×106 mouse melanoma cells were injected into the tail vein. After 2 weeks of feeding, obvious lung metastasis of melanoma cells could be observed without causing the death of the mouse; 3 weeks of feeding, melanoma The number of cell lung metastases is too much, and more than half of the mice died; after 1 week of feeding, the number of melanoma cells metastasized to the lung was small.
Conclusion: Injecting 1×106 mouse melanoma cells through the tail vein and growing for 2 weeks is the recommended method for constructing a C57BL/6J mouse melanoma lung metastasis model.
