【Animal modeling】-DBA/1J mouse collagen induced arthritis model

  Objective: To establish a DBA/1J mouse collagen-induced arthritis model, and to identify the model from morphology, pathology and immunology.

  Method: Mix bovine type Ⅱ collagen (CⅡ) with adjuvant and fully emulsify, subcutaneous injection into DBA/1J mouse tail base for primary immunization, 21 days later, immunize again with the same method. Observe the morphological indicators such as weight change and the degree of joint swelling in mice, conduct pathological examination of the ankle joint, and use CBA method to detect the level of serum cytokines.

  Result: The body weight of the model mice began to drop 35 days after the first vaccination and continued to be lower than that of the control group. On the 28th day, the model mice developed red and swollen feet. The swelling thickness of the feet on the 35th day was significantly higher than that of the control group (P<0.05). By the 62nd day, the incidence rate was as high as 90%. Pathological examination showed that the joint cavity of the model mice was narrowed, the synovial tissue proliferated, inflammatory cells infiltrated, and the cartilage structure was destroyed to varying degrees. Compared with the control group, the inflammatory factors IL-6, IFN-γ, and MCP-1 in the serum of model mice were significantly increased (P <0.05), while="" the="" anti-inflammatory="" factor="" il-10="" was="" slightly="" decreased="" p="">0.05) .

  Conclusion: Subcutaneous injection of bovine type Ⅱ collagen into the tail root of DBA/1J mice can successfully induce CIA model.
