【Animal Modeling】-Nude mice orthotopic transplantation model of liver cancer

  Objective: To establish an orthotopic transplantation model of liver cancer in BALB/c nude mice.

  Method: The experimental animals were general anesthetized by isoflurane vaporization, and the BALB/c nude mice hepatocarcinoma orthotopic xenograft models were established by "embedding method" and "sandwich method" respectively. After the operation, the growth of the tumor tissue was detected with a small animal in vivo imaging system, and histopathological examination of the liver and tumor tissue was performed 4 weeks later.

  Results: The BALB/c nude mouse model of liver cancer orthotopic transplantation established by the sandwich method has the characteristics of short operation time, low difficulty, and high tumor formation rate. The tumor tissue grows well, and there is no widespread tumor implantation in the abdominal cavity. The pathological examination was normal, and the animal mortality was low.

  Conclusion: Compared with the embedding method, the sandwich method has more advantages. It can quickly prepare large quantities of mouse liver cancer orthotopic transplantation models, and provide a more convenient platform for liver cancer in situ pharmacodynamics and other research.
