【Animal Modeling】-Preliminary Study on the Effect of Lipopolysaccharide on Naked Mole Lung Tissue

  Objective To observe and analyze the effects of lipopolysaccharide on the morphological structure and pathological indexes of naked mole rats' lung tissues, and to preliminarily investigate whether the naked mole rats are resistant to LPS.

  Method 10 male naked mole rats were randomly divided into experimental group and control group. The experimental group was intraperitoneally injected with LPS at a dose of 10 mg/kg according to body weight, and the control group was injected with the same volume of 0.9% NaCI solution (normal saline). 6h after the administration, the lungs were collected, and the wet-to-dry weight ratio of lung tissue was measured; making paraffin wax

  section, after HE staining, the microscopic structure of lung tissue was observed under optical microscope; flow cytometry was used to detect the content of reactive oxygen species in lung tissue cells and the rate of apoptosis of lung tissue cells.

  Results The naked mole rats in the experimental group had complete lung tissue structure, and there was no obvious exudate and inflammatory cell infiltration in the alveolar pores. Compared with the control group, the wet-to-dry mass ratio of lung tissue in the experimental group did not change significantly, and the early apoptosis rate did not change significantly, but the late apoptosis rate was significantly increased (P<0.01), and the content of reactive oxygen species was significantly increased (P< 0.05).

  Conclusion Naked mole rats have a certain degree of tolerance to LPS-induced lung injury, and the specific mechanism needs to be further studied.
