【Animal Modeling】-The dose-effect relationship of Xingnaojing injection in the treatment of coma after traumatic brain injury in Beagle dogs

  Objective: To observe the effect of Xingnaojing injection on the dose-effect relationship of Beagle dog coma treatment.

  Methods: 42 male Beagle dogs weighing 8-10kg were randomly divided into normal control group, model control group, Xingnaojing injection dosage group 1 (0.43mL/kg) and dosage group 2 (0.86mL/kg) according to their body weight. , Dose 3 double dosing group ((0.43+0.43) mL/kg), dose 4 group (1.29 mL/kg), dose 5 double dosing group ((0.65+0.65) mL/kg), dogs use Lu Mianning (0.02mL/kg) was anesthetized and fixed in a self-made percussion device. The percussion device was placed at 150cm and hit the dog's head and occipital vertically for 4 consecutive hits to establish a coma model of traumatic brain injury in dogs. Immediately after the model was created, different doses of Xingnaojing injection were given, the administration volume was 10.0mL/kg, the recovery time was observed, and then the brain MRI examination was performed. 14 days after the administration, the MRI examination was performed, and the blood was taken to test the blood routine and liver and kidney Function, serum S-100 calcium binding protein (S-100B), neuron specific enolase (NSE) content. Take the heart, liver, spleen, lung, kidney, adrenal gland, and brain of each group of animals for histopathological examination.

  Result: Xingnaojing injection dose 1, dose 2 (single dose), dose 3 (two doses) can significantly reduce the dog's recovery time. Xingnaojing dose 1, Xingnaojing dose 2, Xingnaojing dose 4 can significantly reduce serum S100B and NSE levels. Xingnaojing dose 1, dose 2, dose 3 (2 doses), dose 4, dose 5 (2 doses) can significantly increase the area of damage recovery from canine brain MRI detection (P<0.05). Xingnaojing injection dose group 1 and 2 can significantly reduce the number of routine blood lymphocytes and eosinophils; Xingnaojing injection dose 1 can significantly reduce the number of canine monocytes; Xingnaojing injection dose 4, 5 pairs There is no obvious influence on the blood routine of dogs. There was no significant difference in liver and kidney function indexes (ALT, AST, TBil, BUN, CRE) in each administration group of Xingnaojing injection. There were no significant differences in the histopathology of the heart, liver, spleen, lung, kidney, and brain organs in each group.

  Conclusion: Xingnaojing injection can significantly improve the degree of coma and brain damage caused by brain trauma in Beagle dogs, and there are no obvious side effects when used in large doses.
