【Animal Modeling】-Efficacy of Eucommia ulmoides extract on mice with mastitis induced by lipopolysaccharide and the way of action

  Objective: To study the therapeutic effect and action route of Eucommia ulmoides extract on mice with mastitis.

  Methods: A mouse model of mastitis was constructed by perfusing LPS through a milk duct. After 12 hours of injection of LPS, eucommia ulmoides with different concentration gradients were administered through the abdominal cavity. The mice were divided into 6 groups. The pathological changes of each group were observed by staining, and the changes of TNF-2α, IL-6, IL-1β cytokines in breast tissue were detected by ELISA method.

  Results: 1) LPS with a concentration of 50 μL 0.2 mg/mL can successfully construct a mouse mastitis model; (2) HE staining results show that Eucommia ulmoides extract can reduce the damage of LPS-induced mastitis in mice. With the injection of Eucommia ulmoides extract The increase in the dose of the solution reduces the damage to the breast tissue, and the injection doses are 20mg/kg and 27mg/kg. The effect is best; (3) The ELISA results show that the extract of Eucommia ulmoides can reduce the inflammatory cytokine IL-1β in the breast tissue. The content of IL-6、TNF-α, and the injection dose is 27mg/kg, the effect is best (P<0.01).

  Conclusion: Eucommia extract can significantly reduce the pathological damage of mouse breast tissue, reduce the release of inflammatory cytokines IL-1β, IL-6, TNF-α in breast tissue, and block neutrophils to breast tissue and breast glands The process of bubble cavity recruitment and migration, thereby reducing breast tissue damage, of which Eucommia ulmoides extract with a concentration of 27mg/kg has the best anti-inflammatory effect.
