Objective: To study the effect of lentivirus-mediated survivin gene specific short hairpin RNA (shRNA) on the growth of human endometrium implanted in the abdominal cavity of nude mice.
Method: Randomly divide 45 nude mice into an experimental group, a negative control group and a blank control group, each with 15 mice. The endometrium of patients with endometriosis was injected into the abdominal cavity of the nude mice, and survivin-shRNA was slowed down at the same time. Virus, empty lentivirus and phosphate buffer were injected into the abdominal cavity of 3 groups of nude mice. After 2 weeks, the success rate of endometrium implantation in each group was observed. The morphological characteristics of ectopic lesions were observed under light microscope. Immunohistochemical method Detect the expression level of survivin in each group of lesions.
Results: The success rate of endometrial implantation in the experimental group (26.67%) was significantly lower than that of the negative control group (73.33%) and the blank control group (80%, P<0.01); the experimental group had dysplasia of living ectopic endometrial glands. The stroma was accompanied by different levels of necrosis; the expression of survivin protein in the lesions of this group also decreased significantly, which was significantly different from the two control groups (P<0.001).
Conclusion: Lentivirus-mediated survivin gene-specific shRNA can significantly inhibit the growth of human endometrium in the abdominal cavity of nude mice by targeting and muting the survivin gene.
