[Animal modeling]-Temozolomide induces caspase-independent programmed death of rat brain glioma C6 cells

  OBJECTIVE: To observe the inducing effect of temozolomide on the programmed death of rat glioma C6 cells and discuss the possible molecular mechanism.

  Method: Observe different concentrations of temozolomide at different time points to treat rat brain glioma rat brain glioma C6 cell line, MTT method observes the inhibition rate and selects the optimal action time and concentration. The cells were treated with 400 μg/ml temozolomide for 24 h, and the induction effect of temozolomide on the programmed death of rat brain glioma C6 cell line was observed by HE staining, Hochest, Western blot, Tunnel, and immunohistochemistry.

  Results: The results of MTT, HE staining, Hochest and Tunnel showed that temozolomide had an apoptosis-inducing effect on rat glioma C6 cells; the results of immunohistochemistry and Western blot showed that the expression level of pro-apoptotic protein Bax was up-regulated, and apoptosis-inhibiting protein Down-regulation of Bcl-2 expression. No major caspase protein expression changes were found.

  Conclusion: Temozolomide has an apoptosis-inducing effect on glioma C6 cells, and this effect may be accomplished through a caspase-independent pathway.
