【Animal Modeling】-Effect of electroacupuncture on the expression of ankle joint myeloid cells in acute gouty arthritis rats to stimulate the expression of receptors

  OBJECTIVE: To discuss the effect of electroacupuncture on the expression of stimulating receptor (TREM)-1 in myeloid cells in the synovial tissue of the tested ankle joint in rats with acute gouty arthritis (AGA).

  Method: 40 SD male rats were randomly divided into normal group, model group, western medicine group, and electroacupuncture group, each with 10 rats; the normal group was fed routinely, and the others were injected with sodium urate solution to establish an AGA model. Two days before modeling, the normal group and model group were given normal saline at 20 ml/kg, the western medicine group was given colchicine solution at 1 mg/kg, and the electroacupuncture group was selected from the test side Sanyinjiao, Xiexi, Kunlun , Frequency 1.5~2 Hz, voltage 9V, current intensity 1~3 mA, density wave, needle retention 20 min, 1 time/d, continuous 9 d. The joint dysfunction of each group of rats was analyzed, and the contents of tumor necrosis factor TNF-α and interleukin IL-1β in the tested ankle joint synovial tissue were detected by ELISA, and the expression of TREM-1 was detected by immunohistochemistry and Western blot. .

  Results: Compared with the normal group, the dysfunction index of rats in the model group was significantly increased (P<0.01), the contents of TNF-α and IL-1β were significantly increased (P<0.05), and the expression of TREM-1 in the tested ankle joint synovial tissue was significant Increase (P<0.05); Compared with the model group, the dysfunction index of rats in the western medicine group and the electroacupuncture group was significant

  Decrease (P<0.01), the contents of TNF-α and IL-1β were significantly decreased (P<0.05), and the expression of TREM-1 in the tested ankle joint synovial tissue was significantly decreased (P<0.05); the="" western="" medicine="" group="" was="" compared="" with="" electroacupuncture="" there="" no="" statistical="" significance="" p="">0.05).

  Conclusion: The mechanism of electroacupuncture treatment of AGA may be through suppression of TREM-1 expression.
