Objective To establish a simple, reliable, and accurate method for noninvasive tracheal intubation in mice inserted into the trachea, which can be used for modeling various mouse lung disease models and drug intervention.
Method Female C57BL/6 mice were injected intraperitoneally with sodium pentobarbital anesthetized, and the mice were suspended through the incisors. Assemble the epidural anesthesia catheter, 1 mL syringe, 10 μL pipette tip, etc. into a set of intubation tools, hold the tool with both hands and sense the mouse trachea, perform non-invasive tracheal intubation through the oral cavity, and give trypan blue or PBS . After the intubation administration, mice in the dye group were observed for the staining of trypan blue in the lung tissue and stomach, and the PBS group was returned to the feeding environment to continue feeding to observe the survival of the mice.
Results There was staining in the lung tissue of the mice in the dye group, and the distribution was relatively uniform, and there was no staining in the stomach. The mice in the PBS group continued to survive after recovery from anesthesia without any abnormalities.
Conclusion A method of using epidural anesthesia catheter, 1 mL syringe, 10 μL pipette tip and other tools for non-invasive endotracheal intubation in mice was established. This method is simple and stable, can accurately determine the insertion of the trachea, has little trauma to animals, and has strong reproducibility.
