Objective: To isolate and expand mouse umbilical cord mesenchymal stem cells to investigate whether they can be induced into cartilage, fat and osteoblasts.
Method: mUCMSCs were isolated, amplified and purified in vitro by adherent culture method, the morphological characteristics of the cells were observed under an inverted microscope, and the expression of antigenic markers of the cells was detected and identified by flow cytometry. The isolated mUCMSCs were induced and cultured into cartilage, adipose and osteoblasts respectively with the induction medium.
Result: The adherent growth of fibroblast-like cells can be isolated from the fresh umbilical cord by the tissue patch culture method. These cells express high CD29, CD90 and CD105, and low express CD34. Axian Blue staining was blue after cartilage induction; oil red O staining after adipogenesis showed red lipid droplets; alizarin staining osteogenic mUCMSC showed red nodules.
Conclusion: mUCMSCs isolated and cultured by adherent culture method can induce differentiation into cartilage, fat and osteoblasts in vitro.
