Objective To establish a purification method for mouse cytomegalovirus to obtain mice without MCMV infection.
Method Using embryo transfer technology, through the use of different hormones, different estrus cycle injection of hormones, recipient mouse strains, different transplantation methods, different stages of embryo transfer, and the number of embryos transferred and other comparative experiments to optimize the purification conditions. The optimized embryo transfer purification method was used to purify the donor mice.
Results The hormone produced by SIGMA, the number of usable embryos obtained by superovulation at a dose of 10IU was about 17; the number of usable embryos obtained by superovulation during the interestrous period of mice was the most, and the number of usable embryos for superovulation was about 23 per mouse. The offspring of C57 male mice mated with ICR female mice were selected as recipient mice, and the birth rate reached 44.5%; fallopian tube transplantation had a better effect than uterine transplantation, the birth rate was 40.64%; the pregnancy rate of transplantation of 2-cell embryos 80%, which is significantly better than single-cell and 8-cell; when the recipient mouse is transplanted with 24 embryos, the litter rate reaches 43.75%. The optimized mouse cytomegalovirus embryo transfer purification method was used to purify mice without MCMV infection.
Conclusion A purification method for mouse cytomegalovirus has been established, which provides a reliable guarantee for obtaining mouse populations without MCMV infection.
