Purpose: The purpose of this experiment is to observe and analyze the expression of thyroid-stimulating hormone-releasing hormone TRH gene in the embryos of Qinghai Lake naked carp at different developmental stages.
Method: Extract the total RNA of Qinghai Lake naked carp to prepare digoxin-labeled antisense TRH gene mRNA probe, and observe the expression of TRH gene by in situ hybridization with a stereo microscope.
Results: Microscopic observation showed that the TRH gene positive hybridization signal appeared in the diencephalon on the 6th to 8th day of embryonic development of Qinghai Lake naked carp; the diencephalon, midbrain, hindbrain and medulla of embryos on the 9th to 11th day appeared in multiple places With positive hybridization signals, the expression level of diencephalon is higher than that of other brain tissues.
Conclusion: In situ hybridization indicated that THR gene was expressed in embryos of Qinghai Lake naked carp at 6-11 days, suggesting that TSH may play an important role in the brain development and maturation of Qinghai Lake naked carp.
