【Animal Modeling】-Directed breeding and identification of Mfge8 knockout homozygous mice and its correlation with autoimmune diseases

  Objective: To breed and identify Mfge8 knockout C57BL/6 mice, obtain homozygous (Mfge8-/-) mice, and compare the serological and histological changes of Mfge8-/- mice.

  Methods: The tails of young mice were cut to extract genomic DNA, and the genotypes of young mice were identified by PCR and agarose gel electrophoresis; the lung tissues of 12-week-old wild-type (WT) and Mfge8-/- mice were detected by TUNEL method, respectively Apoptotic cells in the medium; and immunofluorescence method was used to detect the ANA and AECA levels in the serum of 36-week-old WT and Mfge8-/- mice.

  Results: The constructed knockout mice have been successfully bred and preserved, and homozygous gene deletion mice have been obtained. At the same time, the number of apoptotic cells in the lung tissue of 12-week-old Mfge8-/- mice was significantly more than that of WT mice; The serum of 36-week-old Mfge8-/- mice was positive for ANA and AECA antibodies, while the serum of WT mice was negative.

  Conclusion: After knocking out the Mfge8 gene in C57BL/6 background mice, they are more prone to autoimmune diseases.
