【Animal Modeling】-Comparison of the transfection efficiency of adenoviral vector between tail vein injection and myocardial injection in mice

  Objective: To compare the transfection efficiency of target genes in heart tissue and liver tissue after tail vein injection or myocardial injection of adenovirus vector in mice.

  Method: Construct an adenovirus vector expressing green fluorescent protein GFP (GFP-Ad). Twenty C57BL/6 mice were randomly divided into a tail vein injection adenoviral vector group and a myocardial injection adenoviral vector group with 10 mice each. Real-time quantitative PCR was used to detect the mRNA expression level of GFP in mouse myocardial tissue and liver tissue at different time points. And observe the expression of GFP fluorescence through a fluorescence microscope.

  Result: The expression level of GFP in the heart tissue of the myocardial injection adenovirus vector group was significantly higher than that of the tail vein injection of the adenovirus vector group, and the fluorescence intensity in the heart tissue of the myocardial injection adenovirus vector group was significantly enhanced. At the same time, we found that the mRNA expression level and fluorescence intensity of GFP in the liver tissues of the two groups were significantly higher than those in the heart tissue; and in the tail vein injection of adenovirus vector group, the mRNA expression level and fluorescence intensity of GFP in the liver tissue were at 7 d The peak was reached on the 3rd day in the myocardial injection adenovirus vector group.

  Conclusion: To improve the transfection efficiency of target genes in heart tissue, the method of myocardial injection of adenoviral vector should be adopted; for the transfection efficiency of liver tissue, both injection methods can be used. In view of the less traumatic method of tail vein injection of adenoviral vector, it should be adopted .
