Objective: To provide an animal model for follow-up research by constructing Fndc5 gene knockout mice.
Method: Use TALEN technology to cause deletion mutations in the FNIII domain of Fndc5 gene, and perform genotype identification by sequencing. Establish a stable genetic line by pairing and identify the genotype of the born mice at the mRNA and DNA levels; analyze the weight and blood sugar of the born mice of different ages; determine the Fndc5 in the kidney, liver, brain, muscle, heart and other tissues by qPCR Express the situation.
Result: Four different strains of Fndc5 gene knockout mice were successfully constructed and identified; no significant differences were seen in body weight and blood glucose of born mice of different ages; Fndc5 was confirmed to be highly expressed in muscle, heart and other tissues.
Conclusion: This experiment successfully constructed Fndc5 gene knockout mice internationally, and conducted a preliminary analysis, providing an animal model for in-depth study of the function of Fndc5 gene in vivo.
