【Animal Modeling】-Cloning and molecular characteristics analysis of tree shrew HGF full-length coding sequence

  Objective: To obtain the full-length coding sequence of tree shrew hepatocyte growth factor and analyze its molecular characteristics.

  Method: Using total RNA from tree shrew liver tissue as material, the full-length coding sequence of tree shrew hepatocyte growth factor was obtained by RT-PCR amplification and sequence splicing, and then its sequence and molecular characteristics were analyzed by DNAMAN, Pymol and other bioinformatics software analyze.

  Result: The molecular structure of tree shrew HGF is relatively conserved in evolution, and the overall structure of tree shrew HGF is similar to that of humans. However, tree shrew HGFαNK1 has one more α helix and two longer β-sheets than human HGF. In addition, tree shrew has a strong positive surface charge area in this area. These differences may affect the way tree shrew HGF and its receptors interact.

  Conclusion: This experiment lays the foundation for clarifying the mode of action of tree shrew HGF and its receptor, and the synthesis and preparation of tree shrew HGF.
