Objective To study the effects of Oct4 and Cdx2 knockdown on the expression of genes related to the differentiation of bovine ICM and TE, and to explain the role of Oct4 and Cdx2 in the early development of bovine embryos.
Method RNA interference knocks down the Oct4 or Cdx2 genes of bovine embryos. Real-time fluorescent quantitative PCR and immunofluorescence staining were used to detect the effects of knocking down Oct4 or Cdx2 genes on the early development of bovine embryos and the expression of related genes.
Results Specific siRNA targeting Oct4 or Cdx2 genes can inhibit the expression of Oct4 or Cdx2 in bovine embryos. Oct4 knockdown had no effect on embryo cleavage rate and 8-cell rate, but embryo blastocyst rate was significantly reduced, and mRNA level was significantly decreased (P<0.05). Cdx2 knockdown suppressed Oct4 mRNA expression (P<0.05).
Conclusion Oct4 controls the differentiation of bovine embryos by regulating the expression of Cdx2, Sox2 and Nanog.
