[Animal Modeling]-Based on SDF-1/CXCR4 signal pathway to explore the effect of Xin

Objective Based on the SDF-1/CXCR4 signaling pathway, to explore the effect of Xin'an Biyuan Prescription on the apoptosis of olfactory epithelial neurons in rats with acute sinusitis.

Method 40 SD rats were divided into control group, model group, treatment group and inhibitor group, each with 10 rats. Except for the control group, rats in the other groups were injected with Staphylococcus aureus suspension to construct an acute sinusitis model, and the control group was replaced with sterile normal saline. On the 7th day after the model was established, intragastric administration was used for continuous administration for 1 week, once a day, the treatment group was given Xinan Biyuan Prescription liquid extract calculated according to the weight of the rat (7 g/kg), and the inhibitor group was given 1 mg / kg CXCR4 specific inhibitor AMD3100, the control group and the model group were given the same amount of normal saline. The pathological characteristics of rat sinusitis were observed and scored to identify the establishment of the model. The expression of inflammatory factors TNF-α and IL-1β in the olfactory epithelium of each group was detected by enzyme-linked immunoassay, and each group was detected by TUNEL method. The number of apoptosis in rat olfactory epithelial tissue, immunohistochemical detection of SDF-1 and CXCR4 expression, Western blot was used to detect the pro-apoptotic proteins cleaved caspase 3, Bax and anti-apoptosis in rat olfactory epithelial neuron cells Expression of protein Bcl-2.

Results The pathological score of sinusitis in the model group was significantly higher than that of the control group (P<0.05), and the treatment group was significantly lower than the model group (P<0.05). Compared with the control group, the expression of inflammatory factors in the model group was significantly increased (P<0.05), and the treatment group was significantly lower than the model group (P<0.05). TUNEL staining results show that: compared with the control group, the model group rat olfactory epithelial neuron cell apoptosis significantly increased (P<0.05), the treatment group and the inhibitor group rat olfactory epithelial neuron cell apoptosis was significantly lower In the model group (P<0.05). Compared with the control group, the expression of anti-apoptotic proteins Bcl-2, SDF-1 and CXCR4 in the model group was significantly reduced (P<0.05), and the pro-apoptotic proteins cleaved caspase 3 and Bax increased significantly (P<0.05). ), compared with the model group, the expression of anti-apoptotic proteins Bcl-2, SDF-1 and CXCR4 was significantly increased in the treatment group (P<0.05), and the pro-apoptotic proteins cleaved caspase 3 and Bax were significantly reduced (P<0. 05).

Conclusion Xin'an Biyuan Fangliu extract can improve the apoptosis of supraolfactory neurons in rats with acute sinusitis. The mechanism of action may be closely related to the inhibition of SDF-1/CXCR4 signaling pathway.
