【Animal Modeling】-High-throughput sequencing to study the effects of drinking water with different pH on the intestinal microbes of SPF mice

  OBJECTIVE: To study the effects of drinking water with different pH on gut microbial diversity in SPF mice.

  Methods: Ninety 21-day-old SPF mice were randomly divided into three groups, 30 mice in each group, and were given three types of water, namely acidified water (pH 3.0), neutral water (pH 7.0) and alkaline water (pH 9.0), respectively. and feeding for three months. In each group, 10 animals were sacrificed at 4 weeks, 8 weeks and 12 weeks respectively. The serum was collected to detect bacteria, viruses and parasites according to the SPF national standard project. , construct the library, perform high-throughput (Illumina) sequencing, obtain 10 G data, and perform statistical and cluster analysis on the data.

  RESULTS: Mice treated with pH 3.0 had moderate microbial richness and high stability.

  Conclusion: Under the conditions of large-scale rearing, the selection of pH 3.0 drinking water mice can maintain the diversity of intestinal microorganisms for a long time, and the stability reaches a plateau. This study provides theoretical and technical support for the production practice of long-term and stable maintenance of animal quality, and is also a specific application of gastrointestinal metagenomics technology in the field of animal quality control.
