Objective: The osteoporosis model constructed by ovariectomized rats is the most suitable model for the study of postmenopausal osteoporosis. However, there is little research on the effect of ovariectomized rats on the skin. The purpose of this study was to observe the effect of ovariectomy on skin aging indicators in rats.
Methods: Twenty 3-month-old healthy SD female rats were randomly divided into a sham operation group (Sham group) and an ovariectomy group (OVX group). Three months after the ovaries were removed, serum estradiol was measured, and serum estradiol was measured. Aging-related biochemical indicators such as catalase (CAT), glutathione peroxidase (GSH-Px), superoxide dismutase (SOD) activity and malondialdehyde (MDA), hydroxyproline ( HYP) and lipofuscin (LF) content, and observed histopathological changes in the skin, and quantitatively analyzed with a computer image analysis system.
Results: Serum estradiol level of rats in OVX group was significantly decreased; histopathological observation showed that the thickness of epidermis and dermis in OVX group was significantly thinner than that in Sham group (P<0.01), and the collagen fibers in the skin were reduced and loosely arranged; , the activities of CAT, GSH-Px and SOD in the skin of the OVX group were significantly decreased (P<0.01), the content of MDA was significantly increased (P<0.01), the content of HYP in the skin tissue was significantly decreased, and the content of LF was significantly increased (P<0.01). 0.01).
Conclusion: 3-month-old rats can induce skin aging 3 months after ovariectomy, which may be a suitable model for the study of postmenopausal skin aging.
