Objective: To investigate the effect of recombinant human erythropoietin (rhEPO) on the expression of brain-derived neurotrophic factor (BDNF) in different parts of brain tissue of aging rats.
Methods: Forty 2-month-old male SD rats were randomly divided into 4 groups: negative control group (N), D-galactose group (D), EPO intervention group (E), positive control group (P), each group 10. The changes of BDNF expression in different parts of the brain tissue of D-galactose-senescent rats after exogenous rhEPO intervention were observed by immunohistochemical staining.
RESULTS: Comparing different groups of rats, it was found that there were significant differences in the expression of BDNF at the same site: the counts of BDNF-positive cells in the hippocampus CA1, CA3, DG and frontal cortex of rats in group D were significantly lower than those in the same site in group N (P<0.05). After rhEPO intervention, the counts of BDNF positive cells in hippocampus CA1, CA3, DG and cerebral cortex motor area of rats in group E and group P were significantly increased compared with those in group D and group N (P<0.05); difference. The comparison of BDNF-positive cells in different parts of the rats in the same group showed that the number of BDNF-positive cells in different parts of the rats in the same group was significantly different (P<0.05). area, hippocampal CA1 area.
Conclusion: rhEPO is universal in enhancing the expression of BDNF in rat nerve cells, suggesting that rhEPO may play a protective effect on nervous system aging by enhancing endogenous BDNF.
