Objective: To compare the differences of glutamate-induced cell swelling by cultured astrocytes (AST) in Wistar rats and SD rats.
METHODS: Wistar rats and SD rats were used for AST primary and subculture on 1 d of neonatal period. On 10 d of subculture, they were given 1 and 10 mmol/L glutamate to incubate for 48 h, and the cells were detected by the release rate of lactate dehydrogenase. Activity, glial fibrillary acidic protein (GFAP) immunofluorescence staining, Image Pro Plus software to measure cell perimeter to indicate cell volume changes, and reverse transcription real-time fluorescent quantitative PCR to detect aquaporin 4 (AQP4) mRNA expression changes.
RESULTS: There was no difference in the effect of glutamate on the AST activity of different breeds of rats (P>0.05). Under normal conditions, the AST circumference of Wistar rats was smaller than that of SD rats, and after glutamate treatment, it was significantly larger than that of SD rats (P<0.05). After administration of 1 mmol/L glutamate, the AQP4 mRNA expression in AST in Wistar rats was significantly higher than that in SD rats (P<0.05).
Conclusion: The glutamate-induced cell swelling in Wistar rats is more obvious than that in SD rats, and the effect of glutamate on the expression of AQP4 on AST in the two varieties of rats is different.
