Objective: To discuss the effect of different doses of propofol on the expression of MTA1 and Wnt1 in rat MADB106 cells lung metastasis and tumor tissue.
Methods: 40 Fischer344 male rats were randomly divided into: normal saline group (S group); fat emulsion group (F group); propofol 30 mg/kg group (P30 group) and 50 mg/kg group (P50 group) ), 10 in each group. 1% pentobarbital sodium 50 mg/kg was intraperitoneally injected into the same volume of the above drugs through the femoral vein, and 0.5 ml of MADB106 tumor cells (2 × 105 cells) were intravenously injected 1 hour later. After 3 weeks, the mice were sacrificed, and the number of lung metastases was counted; the expression of MTA1 and Wnt1 in lung tumor tissue was detected by immunohistochemistry, and quantitatively analyzed by IPP software.
RESULTS: There was no significant difference in the number of lung metastases and the expressions of MTA1 and Wnt1 in the tumor tissue between the F group and the S group (P>0.05). Compared with the S group, the number of lung metastases and the expressions of MTA1 and Wnt1 in the tumor tissues of the P30 and P50 groups were significantly decreased (P<0.01). Correlation, Pearson correlation coefficients were -0.879, -0.980 and -0.916 (P<0.01). The expressions of MTA1 and Wnt1 were positively correlated, and the Pearson correlation coefficient was 0.902 (P<0.01).
Conclusion: Propofol inhibits lung metastasis of MADB106 tumor cells in a dose-dependent manner, and down-regulates the expressions of MTA1 and Wnt1 in metastatic tumors.
