Objective: To investigate the expression of canonical transient receptor potential channel 1 (TRPC1) in mouse lung tissue under ozone exposure.
Methods: The mouse lung tissue inflammation model was established by ozone exposure. After the model was successfully established by the behavioral observation of mice, cell count of bronchoalveolar lavage fluid and lung tissue pathological morphological analysis, RT-PCR method was used to detect TRPC1 in lung tissue. mRNA expression, Western blot and immunohistochemical staining were used to observe the expression and localization of TRPC1 protein in lung tissue.
Results: RT-PCR and Western blot results showed that compared with the control group, the expression of TRPC1 mRNA and protein in the lung tissue of the mice in the ozone group was up-regulated (P<0.05). The expression of TRPC1 protein in alveolar epithelial cells, bronchial epithelial cells, and infiltrating inflammatory cells was significantly enhanced compared with the control group (P<0.05). The total number of leukocytes, macrophages, neutrophils and lymphocytes in the washing solution were significantly positively correlated (P<0.01).
Conclusion: TRPC1 may be involved in the pathogenesis of lung inflammation in mice.
