【Animal modeling】-Effect of Helicobacter hepatica infection on dextran sulfate sodium-induced chronic colitis model in mice

  Objective To investigate the effect of Helicobacter hepatica infection on chronic colitis induced by sodium dextran sulfate in mice.

  Methods Forty male IL-10* mice were randomly divided into 4 groups, namely control group, H.h group, DSS group and H.h+DSS group. H.h group and H.h+DSS group were infected with Hh by gavage. Five days after Hh infection, mice in DSS group and H,h+DSS group drank 2% DSS water continuously for 7 d, then returned to normal drinking water for 5 d, and then drank 2% DSS water continuously for 7 d; the other groups drank normal water. After the experiment, the mice in each group were sacrificed, and the colon was taken and its length was measured, and then divided into 3 parts for histopathological examination, cytokine mRNA transcription level detection and protein expression level detection.

  Results The results of pathological examination showed that compared with the DSS group, the H, h+DSS group mice had increased lymphocyte infiltration, severe epithelial loss, aggravated fibrosis, obvious crypt atrophy, and the inflammatory cytokine interleukin-6. and tumor necrosis factor alpha transcription levels were up-regulated, and signal transducer and activator of transcription 3 protein expression levels were elevated.

  Conclusion H.h infection exacerbates DSS-induced colitis in mice.
