Objective To observe the effects of Tanreqing injection on the cardiovascular system, respiratory system and central nervous system of ICR mice in Beagle dogs, and to provide safety experimental data for its clinical use.
Methods Twenty-four Beagle dogs, half male and half female, were randomly divided into low-dose (2.5 mL stock solution/kg), medium-dose (5.0 mL stock solution/kg), and high-dose (10.0 mL stock solution/kg) Tanreqing injection groups according to body weight. and the solvent control group, 6 in each group; under anesthesia, the right forelimb medial subcutaneous cephalic vein was injected by bolus. Blood pressure, heart rate, electrocardiogram and respiration at 60, 90, 120, 150, 180, 210 and 240 min. 160 ICR mice were divided into half male and female, 60 of which were randomly divided into low-dose (3.6 mL stock solution/kg), medium-dose (6.25 mL stock solution/kg), and high-dose (12.5 mL stock solution/kg) Tanreqing according to body weight. The general behavior and white hair activity of the mice in the injection group, the solvent control and the positive control group were observed after administration by tail vein injection; for 50 mice in the same group, the coordinated movement at different time points after administration was measured; In 50 mice in the same group, the synergistic effect of sodium pentobarbital sodium in subthreshold sleep after administration was determined.
Results There were no significant differences in the systolic blood pressure, mean arterial pressure, electrocardiogram and respiratory rate of Beagle dogs between each dose group of Tanreqing injection and the solvent control group at the same time point and before self-administration (P>0.05). ), there were statistically significant differences in diastolic blood pressure and heart rate at individual time points (P<0.05 or P<0.01), but no significant dose or time-effect relationship was found. After administration, the spontaneous activity of ICR mice in the high-dose group decreased at each time point (P<0.05, P<0.01 or P<0.001), and the coordinated motor scores were significantly increased at 30, 50 and 90 minutes after administration (P<0.05, P<0.01 or P<0.001). <0.05, P<0.01 or P<0.001).
Conclusion Under the experimental conditions, a single intravenous injection of Tanreqing injection has no obvious effect on the cardiovascular and respiratory systems of Beagle dogs; high-dose Tanreqing injection has a certain effect on the central nervous system of ICR mice, mainly It is manifested as decreased spontaneous activity and impaired coordination of movement, which may be related to its main active ingredient, bear bile powder.
