[Operation steps] Choose 300~500g adult male guinea pigs, use normal saline containing 0.25% calf serum albumin to dilute the platelet active factor to 500μg/ml, and inhale at a dose of 1500μg/kg, which can cause guinea pig asthma attack.
[Result analysis] Platelet active factor (PAF) is the only inflammatory mediator known to cause airway hyperresponsiveness. PAF may cause asthma attacks due to PAF chemotaxis through the activation of eosinophils, degranulation, release of eosinophil protein X (Epx), eosinophil cationic protein (ECP) and basic protein and other cytotoxic substances causing air Trau epithelial cells are damaged and shed. The activated eosinophils themselves synthesize and release PAF, which aggravates this process and eventually causes airway hyperresponsiveness. Platelet active factor triggers guinea pig asthma attack without sensitization process, and directly uses its characteristics to trigger airway hyperresponsiveness. This model is mainly used to study the etiology and pathogenesis of asthma. Nebulized inhalation equipment is required to strictly control the inhalation concentration.
