  【Steps】

  (1) Acute renal failure model caused by the clamping of the renal artery in a dog: Take a male dog of 10 to 20 kg and anesthetize it with 30 mg/kg sodium pentobarbital, surgically expose the left renal artery to be clamped and relax for 1 hour , To reperfuse the blood flow, collect urine from the side ureter, and perform phenotypic analysis of acute renal failure.

  (2) Rat model of acute renal failure induced by surgery: SD male rat, weighing 280~350g, anesthetized with pentobarbital sodium 50mg/kg, cut both sides of the abdomen, remove the right kidney, separate the left kidney pedicle, and expose the left kidney The artery and vascular clip were clamped for 60 minutes, then the wound was sutured, and the animal was recovered. Blood creatinine was collected every day after tail docking, and the phenotype analysis of acute renal failure was performed.

  (3) Acute renal failure model caused by norepinephrine in domestic dogs: Take male dogs of 10-20 kg and anaesthetize them by intravenous injection of sodium pentobarbital 30 mg/kg, then insert a catheter into one ureter to collect urine. Norepinephrine was instilled in the other renal artery at a rate of 0.75 mg/min. The drug was stopped after 40 minutes, and the phenotype analysis of acute renal failure could be performed within 1 to 4 hours.

  (4) Rat hemoglobinuria acute renal failure model: Take male rats weighing 150~250g, place them in a metabolic cage, record normal drinking water and urine output, keep water for 24 hours before medication, and select lighter and dehydrated rats , 50% glycerol was injected into the muscles of both hindlimbs of rats at 10ml/kg. After the injection, the rats eat and drink freely. Proteinuria appeared after 2h, a stable model was formed after 48h, and the experiment could be carried out in 2~5d.

  (5) Acute renal failure model caused by glycerol: Take male rats weighing 150-250g and inject 50% glycerol 10ml/kg once intramuscularly. After 24 hours, there will be hematuria, increased urea nitrogen, oliguria or anuria, similar to acute kidney decline.

  【Result analysis】Urine, blood creatinine, serum total protein and albumin, serum urea nitrogen, serum sodium and potassium, and renal pathological examinations are routinely performed. The renal failure caused by the acute renal failure model caused by norepinephrine in dogs is reversible. If the infusion is delayed for 2 hours, it can cause extensive renal parenchymal necrosis and acute tubular necrosis. If the blood flow is blocked for more than 4 hours, the rat will die of uremia. This model can be used to observe the effect of drugs on acute renal failure.
