【Animal modeling】-Protective effect of Guanxinning tablet on myocardial ischemia-reperfusion injury in rats

  Objective: To observe the effect of Guanxinning Tablets on myocardial infarction and cardiac autonomic function in rats with myocardial ischemia-reperfusion injury (MI/RI).

  Methods: 70 SD rats were randomly divided into 7 groups, namely sham operation group, MI/RI group, GXNT 75 mg/kg, 150 mg/kg, 300 mg/kg, 600 mg/kg dose group and 300 mg/kg dose group. kg Compound Danshen Tablets (DST) group, 10 in each group. All rats were given prophylactic oral gavage for 7 days, and then the left anterior descending coronary artery was ligated to establish MI/RI rat model. During this period, rat electrocardiograms were recorded and analyzed for ECG J-point and heart rate variability (HRV) indicators, and the myocardium was double stained with Evans blue (EB) and 2,3,5-triphenyltetrazolium chloride (TTC) and analyzed. The myocardial infarction area and the pathological changes of the myocardium were observed by HE staining, and the levels of plasma lactate dehydrogenase (LDH), creatine kinase (CK), malondialdehyde (MDA) and nitric oxide (NO) were determined.

  Results: Compared with MI/RI group, Guanxinning tablet group and Fufang Danshen tablet group could significantly reduce myocardial infarction size and inhibit the increase of LDH and CK activity (P<0.05, P<0.01), and could reduce the reperfusion period The ∑-J value and the average value of J point (P<0.05, P<0.01) in rats significantly increased HRV and plasma NO levels and decreased plasma MDA content (P<0.05, P<0.01), and could improve myocardial histopathology.

  Conclusion: Guanxinning Tablets can alleviate myocardial infarction in MI/RI rats and improve cardiac autonomic function.
