【Animal Modeling】-Effect of continuous multiple application of propofol and vitamin C on anesthesia in mice

Objective: To investigate the anesthesia effect of propofol or propofol combined with vitamin C on mice for 6 consecutive days.

Methods: 40 Kunming rats were randomly divided into propofol 80 mg/kg group (P80 group), propofol 70 mg/kg+vitamin C50 mg/kg group (P70+Vc50 group), propofol 55 mg/kg+ Vitamin C100 mg/kg group (P55+Vc100 group), propofol 50 mg/kg+vitamin C200 mg/kg group (P50+Vc200 group), a total of 4 groups, 10 mice in each group, intraperitoneally injected In the same way, the mice were administered at the same time point for 6 consecutive days, and the anesthesia induction time and maintenance time of the mice were observed and recorded, and compared.

RESULTS: Compared with the propofol group (P80) on the same day, the anesthesia maintenance time of the P55+Vc100 group and the P50+Vc200 group on the 1st, 2nd, and 3rd day of administration was shortened (P<0.05). Compared with the anesthesia maintenance time on the 1st day in the same group, the remaining 5-day anesthesia maintenance time of the mice in each group was shortened (P<0.01). The maintenance time was shortened (P<0.01), and the maintenance time of anesthesia was shortened on the 4th, 5th, and 6th days in each group (P<0.01). The incidence of righting reflex disappearance in each group gradually decreased from day 1 to day 6, and the trend in each group was close to the same.

Conclusion: The compatibility of vitamin C and propofol can reduce the dose of propofol to a certain extent on the premise of ensuring the anesthetic effect, and at the same time can reduce the occurrence of propofol tolerance. reaction.
