Objective: To investigate the cardioprotective effect and mechanism of Zhenwu Decoction on uremic cardiomyopathy in mice.
Methods: The uremic cardiomyopathy model was established by most nephrectomy. C57BL/6 mice were randomly divided into 3 groups: sham operation group, model group and Zhenwu decoction group. The mice in the Zhenwu decoction group were given Zhenwu decoction by gavage after surgery, and the mice in the sham-operated group and the model group were given the same amount of distilled water by gavage after surgery. Cardiac color Doppler ultrasound, HE staining of cardiac tissue and blood biochemistry were used to observe the changes of cardiac function, structure and renal function in uremic cardiomyopathy mice, and Western blotting was used to detect the expression of p-AMPK and p-mTOR in cardiac tissue.
Results: Compared with the sham operation group, the body weight of the mice in the model group decreased significantly (P<0.05), the cardiac mass and cardiac index increased significantly (P<0.05), and the thickness of the left ventricular posterior wall increased during the diastolic and systolic periods (P<0.05). ), HE staining showed myocardial cell hypertrophy, blood urea nitrogen and creatinine were significantly increased (P<0.05). Compared with the model group, the heart mass and cardiac index of the mice in the Zhenwu decoction group decreased (P<0.05), the thickness of the posterior wall of the left ventricle during diastole and systole decreased (P<0.05), and the pathological changes were alleviated. Compared with the sham-operated group, the expression of p-AMPK in the model group was significantly decreased, and the expression of p-mTOR was significantly increased (P<0.05). The expression of mTOR was significantly decreased (P<0.05).
Conclusion: Zhenwu Decoction inhibits myocardial hypertrophy in mice with uremic cardiomyopathy, and this effect may be achieved through the AMPK-mTOR signaling pathway.
