Objective: To investigate the protective effect of Src kinase inhibitor PP2 on hypoxia/reoxygenation injury of rat astrocytes.
Methods: The experiment was divided into normal control group, hypoxia/reoxygenation group (H/R; hypoxia 8 h/reoxygenation 24 h), PP2 + hypoxia/reoxygenation group (PP2+H/R; given before hypoxia) 10 μmol/L PP2 for 24 h). The survival rate of astrocytes in each group was detected by MTT method, the apoptosis rate of astrocytes in each group was determined by flow cytometry, and the expression of Src kinase, Bax and Bcl-2 in astrocytes in each group was detected by Western blotting method. Results: The results of MTT assay showed that the cell viability decreased after H/R injury, while the cell viability increased after pretreatment with PP2 (P<0.01). Flow cytometry results showed that cell apoptosis increased after H/R injury. However, apoptosis was significantly decreased after pretreatment with PP2 (P<0.01). The results of Western blotting showed that the expression of Src kinase increased after H/R injury, while the expression of Src kinase decreased after pretreatment with PP2. The ratio of apoptosis-related protein Bax/Bcl-2 increased after H/R injury, and the ratio of Bax/Bcl-2 decreased significantly after pretreatment with PP2 (P<0.01).
Conclusion: PP2 can protect astrocytes from H/R injury, and the mechanism may be related to inhibiting the protein expression of Src kinase and activating the anti-apoptotic mechanism.
