Objective To study the effects of ambient air exposure on microorganisms and metabolites in prepared fecal suspensions.
Methods The feces of specific pathogen-free Bama miniature pigs were collected: (1) untreated group (C group); (2) aerobic exposure treatment group (T1 group); (3) anaerobic exposure treatment group (T2 group). The microbial community composition and metabolites of fecal samples in each group were compared using amplicon sequencing technology and untargeted metabolome.
Results (1) The preparation of fecal suspension in both groups T1 and T2 preserved all bacterial phyla in fresh feces (group C), mainly Firmicutes and Bacteroidetes, but significantly reduced Treponema bacteria. (2) Ascomycota and Basidiomycota fungi were mainly in pig feces, and the preparation of fecal suspension significantly increased the abundance of Ascomycota fungi in pig feces; aerobic exposure treatment removed nodules in pig feces. A single fungus. (3) LC-MS results showed that 402 and 195 metabolites were identified in the positive and negative ion modes, of which 155 and 201 were differential metabolites in the T1:C metabolic set and T2:C metabolic set, respectively.
Conclusion Although the preparation time of pig manure suspension is not long, the effect of aerobic exposure on the abundance of fungal communities in the samples is higher than that of anaerobic exposure, both of which significantly reduce the abundance of harmful flora and related metabolites such as Treponema and Spirochaetes, and increase the security of FMT.
