OBJECTIVE: To establish a genetic detection method for pigeon microsatellite DNA for closed colony experiments.
Methods: A total of 61 pigeon microsatellite loci were selected and designed through literature search and SSRHunter software screening. The pigeon genome samples were used for PCR amplification and condition optimization. The results of agarose gel electrophoresis were used for preliminary screening to obtain alleles. There are 20 microsatellite loci with abundant and clear amplified bands. Through the comparative analysis of STR scanning, a total of 16 loci suitable for genetic quality detection of experimental pigeons were selected and applied to two closed groups of pigeons.
RESULTS: Using the screened 16 microsatellite DNA loci, a genetic detection method for pigeons used in closed group experiments was preliminarily established.
Conclusion: The genetic detection method of pigeons used in closed group experiments based on microsatellite DNA was initially established, which laid the foundation for the genetic quality control of pigeon populations used in closed group experiments.
