Objective: To establish a genetic detection method of chicken microsatellite DNA for experimental use.
Methods: 72 microsatellite loci were preliminarily screened through literature search and analysis, and PCR and STR scanning were used to determine the loci with good amplification effect and strong stability. The loci suitable for genetic testing of chickens for closed group and haplotype experiments were preliminarily determined. Point combination, and applied in groups of 3 closed-group experimental chickens and 3 haplotype experimental chickens.
RESULTS: 28 microsatellite loci combinations suitable for genetic testing of chickens used in closed flock experiments and 14 microsatellite loci combinations suitable for genetic testing of chickens used in haplotype experiments were preliminarily determined.
Conclusion: The genetic detection method of experimental chickens has been preliminarily established.
