【Animal modeling】-phenotype and pathological observation of leptin receptor gene knockout SD rats

Objective: To observe various parameters of the leptin receptor gene knockout rat model of obesity, metabolic syndrome and prediabetes established by CRISPR/Cas9 technology, and to provide a more ideal basis for the molecular mechanism research of metabolic diseases and the development of new drugs. animal model.

Methods: 1) The body weight, food intake, random blood glucose and fasting blood glucose of Lepr-/- rats were measured regularly; glucose tolerance test and insulin tolerance test; serum lipid parameters; 2) Lepr-/- rats were observed for 14 and 18 weeks Pathological changes of major organs; 3) Observe the lipid deposition in organs.

Results: Lepr-/- rats showed obvious obesity, high food intake, impaired glucose tolerance, insulin resistance and dyslipidemia; pancreatic islet hyperplasia and cardiac myocyte hypertrophy, fatty liver, obesity-related nephropathy at 14 and 18 weeks; Cells, renal tubular epithelial cells and lipid deposition in skeletal muscle fibers.

Conclusion: Lepr-/- rats are ideal animal models for the study of the etiology of metabolic diseases and the development of new drugs.
