【Animal modeling】-Determination of fecal microbial diversity of common cotton-eared marmosets based on high-throughput sequencing technology

  Objective: To analyze the structure and composition of the fecal flora of the common cotton-eared marmoset by using high-throughput sequencing technology, and to lay a foundation for the further development and utilization of new experimental animals.

  Methods: The feces of 4 adult male common cotton-eared marmosets were collected, and the V3-V4 region was amplified with bacterial 16S rRNA universal primers. The IlluminaMiSeq sequencing platform was used to study the common cotton-eared marmoset fecal microorganisms.

  RESULTS: A total of 315,511 valid sequences and 596 OTUs were obtained by sequencing. A total of 9 phyla, 14 classes, 26 orders, 50 families, 82 genera and 64 species and There are 226 OTUs. Among them, 1) the dominant phyla are Bacteroidetes and Firmicutes, with an average content of 54.52% and 25.39%, respectively 2) The most abundant classes are Bacteroidia and Firmicutes. Negativicutes, with an average content of 54.5% and 17%. 3) Lactobacillales and Bacteroidales have the highest abundance, 50.01% and 20.52%. 4) Prevotella Prevotellaceae and Bifidobacteriaceae account for the highest abundance, with an average content of 43.14% and 11.33%, respectively. 5) The abundance of Bifidobacterium and Bacteroides is higher, The average content was 11.33% and 11.12%, respectively. 6) The beneficial bacteria Bifidobacterium is abundant, but it is contained in the tested samples. 7) The top 10 species with the highest abundance are classified into 7 families and 5 classes. These 10 species account for 33.16%, and the total of the other 53 species only accounts for 0.74%. Other species have not been identified, and their relative abundance is still relatively high, and further research is needed. 8) PICRUSt function prediction analysis: amino acids Metabolic functions such as transport and protein translation, folding and genetic information processing functions are abundant.

  Conclusion: Using high-throughput sequencing technology, the fecal flora of the common cotton-eared marmoset has been comprehensively detected. The common cotton-eared marmoset fecal bacterial composition is rich in diversity, and many of them have not been classified and identified with relatively high relative abundance. High bacteria need further study.
