【Animal Modeling】-Oriented breeding and identification of Mfge8 knockout homozygous mice and their correlation with autoimmune diseases

  Objective: To breed and identify Mfge8 knockout C57BL/6 mice, obtain homozygous (Mfge8-/-) mice, and compare the serological and histological changes of Mfge8-/- mice.

  Methods: Genomic DNA was extracted from the tail of young mice, and the genotypes of young mice were identified by PCR and agarose gel electrophoresis. The lung tissues of 12-week-old wild-type (WT) and Mfge8-/- mice were detected by TUNEL method, respectively. and the contents of ANA and AECA in serum of 36-week-old WT and Mfge8-/- mice were detected by immunofluorescence method, respectively.

  Results: The constructed knockout mice were successfully bred and conserved, and homozygous gene deletion mice were obtained. At the same time, the number of apoptotic cells in the lung tissue of 12-week-old Mfge8-/- mice was significantly higher than that of WT mice; The serum of 36-week-old Mfge8-/- mice was positive for ANA antibody and AECA antibody, while the serum of WT mice was negative.

  Conclusion: Mfge8 gene knockout in C57BL/6 background mice is more prone to autoimmune diseases.
