Objective: To investigate the effect of P-selectin gene (P-selectin) deletion on the physiological characteristics of mice.
Methods: P-selectin knockout (PKO) mice and C57 mice were raised in an SPF environment, and they were identified as pure and PKO mice by genetic identification. The activity and breeding status of PKO mice were observed, and blood routine was detected to determine the effect of P-selectin knockout on the general condition of mice. Mice were sacrificed at 6 weeks and 18 weeks, respectively, and the effect of P-selectin knockout on the structure of various organs of mice was observed by eosin & hematoxylin (HE) staining.
RESULTS: The breeding status of PKO mice was similar to that of normal C57 mice, and the blood routine was normal. HE staining showed that the structures of liver, spleen, lung and kidney of PKO mice were normal.
Conclusion: The deletion of P-selectin does not affect the reproduction and blood of mice, nor does it have obvious effects on important tissues and organs. It provides an animal model for further research on the role of P-selectin in diseases. theoretical basis.
