Objective: To observe and analyze the differences in reproductive system structure and function between fertile and infertile male naked mole-rats, and preliminarily explore the reasons why male infertile individuals cannot reproduce.
METHODS: Five male naked mole rats were fertile and infertile. One testis and epididymis were collected first, and the organ coefficients were converted after weighing. The other testis was collected and fixed with neutral formaldehyde solution. Tissue sections were prepared; the other side of the epididymis was taken for the determination of sperm count and sperm motility; serum was collected to measure the concentrations of luteinizing hormone (LH) and testosterone (T) in serum.
Results: Compared with the fertile individual group, the testicular weight of the infertile individual group decreased (P < 0.05), the weight of the epididymis decreased significantly (P < 0.01), and the testicular coefficient and epididymal coefficient were significantly decreased (P < 0.01); sperm The number of spermatozoa decreased significantly (P < 0.01), and the motility of sperm was also significantly weakened (P < 0.01); the levels of serum luteinizing hormone and testosterone were significantly decreased (P < 0.01); the observation of testicular tissue sections showed that spermatogenesis at all levels of infertile individuals was significantly lower (P < 0.01). The cells were severely shed, the arrangement was disordered, and the Sertoli cells and stromal cells were reduced.
Conclusion: The reproductive system of male naked mole-rats infertile individuals has atrophy of testis and epididymis, decreased spermatogenesis and decreased secretion of sex hormones.
