OBJECTIVE: To provide an animal model for subsequent research by constructing Fndc5 knockout mice.
Methods: Deletion mutations were created in the FNIII domain of the Fndc5 gene using TALEN technology, and the genotype was identified by sequencing. Establish stable genetic lines by pairing and identify the genotype of newborn mice at the mRNA and DNA levels; analyze the body weight and blood sugar of newborn mice at different ages; determine the expression of Fndc5 in kidney, liver, brain, muscle, heart and other tissues by qPCR express the situation.
RESULTS: Four different strains of Fndc5 knockout mice were successfully constructed and identified; there was no significant difference in body weight and blood glucose of mice born at different ages; it was confirmed that Fndc5 was highly expressed in muscle, heart and other tissues.
Conclusion: This experiment successfully constructed Fndc5 knockout mice in the world, and carried out preliminary analysis, which provided an animal model for in-depth study of the function of Fndc5 gene in vivo.
