【Animal Modeling】-Cloning and Molecular Characterization of the Full-length Coding Sequence of Tree Shrews HGF

  Objective: To obtain the full-length coding sequence of tree shrew hepatocyte growth factor and analyze its molecular characteristics.

  Methods: Using total RNA of tree shrew liver tissue as material, the full-length coding sequence of tree shrew hepatocyte growth factor was obtained by RT-PCR amplification and sequence splicing, and then its sequence and molecular characteristics were analyzed by bioinformatics software such as DNAMAN and Pymol. analyze.

  RESULTS: The molecular structure of tree shrew HGF was relatively conserved in evolution, and the overall structure of tree shrew HGF was similar to that of humans. However, compared with human HGF, the HGFαNK1 region of tree shrew has one more α-helix and two longer β-sheets. In addition, tree shrew has a strong positive surface charge region in this region. These differences may have implications for the mode of action of tree shrew HGF and its receptors.

  Conclusion: This experiment lays a foundation for clarifying the mode of action of tree shrew HGF and its receptors and for the synthesis and preparation of tree shrew HGF.
