Objective To explore the protective effect and mechanism of quercetin on cadmium-induced liver injury in mice.
Methods C57BL/6J mice were divided into control group, cadmium group (gavage of cadmium chloride), quercetin group (gavage of quercetin), cadmium and quercetin co-treatment group (gavage of cadmium chloride + quercetin) White). After the administration, the body weight of mice, the activities of aspartate aminotransferase (AST) and alanine aminotransferase (ALT) in serum, and the total serum and liver tissue levels were determined.
The contents of cholesterol (TC) and triacylglycerol (TG); the pathological changes of liver tissue were observed by HE staining, and the changes of lipid droplets in liver tissue were observed by oil red O staining; Western blotting was used to detect the key enzymes of lipid synthesis, fatty acid synthase (FAS) and its decomposition. Changes in the key enzyme lipoprotein lipase (LPL).
Results Compared with the control group, the body weight of cadmium-exposed mice decreased significantly (P < 0.01), the activities of serum ALT and AST increased significantly (P < 0.01), and the liver tissue had obvious pathological changes of liver injury. The contents of TC and TG were significantly increased (P < 0.01), and a large number of fat droplets were deposited in the liver tissue (P < 0.01).
adjustment (P<0.01). Quercetin intervention can significantly improve the abnormal changes of the above indicators (P<0.05 or P<0.01).
Conclusion Quercetin can play a protective effect on cadmium-induced liver injury in mice by regulating the disorder of liver lipid metabolism.
