【Animal Model】-Long-term effects of sublethal dose of γ-ray whole body irradiation on hematopoietic immune function in mice

  Objective: To observe the changes of hematopoietic and immune function indexes in C57BL/6J mice 6 months after total body irradiation with 4Gy and 6Gy.

  METHODS: C57BL/6J mice aged 8-12 weeks were irradiated with 4Gy and 6Gy whole body respectively. After 6 months of irradiation, peripheral blood cell count and classification, thymus coefficient and spleen coefficient, peripheral blood and spleen immune cell typing, and spleen T were detected. Cellular p16 expression level.

  Results: 6 months after irradiation, the white blood cell and red blood cell counts in peripheral blood were still significantly lower than those in the control group. There was no significant difference between platelets and the control group. Compared with the control group, the thymus coefficient was significantly decreased 6 months after 6Gy irradiation (P<0.05). There was no difference in the spleen coefficient. Analysis of peripheral blood flow showed that after 4Gy and 6Gy irradiation, B Lymphocytes and NK cells were still lower than those in the control group, indicating incomplete recovery after 6 months of exposure. The CD4 ratio in the exposure group was higher than that in the control group. There was no difference between CD8 and monocytes and the control group. The spleen immune cells were analyzed by flow cytometry Analysis, the results showed that 6 months after 4Gy and 6Gy irradiation, B220 was significantly decreased in the control group, with extremely significant differences (P<0.01). Compared with the control group, the expression of p16 in spleen T cells in the 6Gy group increased trend, but no significant difference.

  CONCLUSION: After sub-lethal dose irradiation, all tests basically recovered, but the neutrophils in peripheral blood continued to increase, and the thymus coefficient and the proportion of B lymphocytes in peripheral blood and spleen were still lower than those in the control group. This suggests that there may be long-term damage. . This experimental data provides basic data for the long-term damage of hematopoietic immune function after medium and high dose irradiation.
