Objective: To observe the damage effect of X-ray whole body irradiation on the function of hematopoietic immune system in KKAy mice, a model of type 2 diabetes mellitus, and compare it with control C57 mice.
Methods: KKAy mice were divided into a control group and an irradiation group. The mice in the irradiation group were irradiated with X-rays at a dose of 4 Gy, and C57 mice were used as controls. The peripheral blood of the mice was detected 15 days after irradiation, and flow cytometry was performed. The proportion of hematopoietic progenitor cells, hematopoietic stem cells and long-term hematopoietic stem cells in the bone marrow, the proportion of B cells and T cells in the spleen, the proportion of CD4CD8 double positive T cells, CD4 single positive T cells and CD8 single positive T cells in the thymus. By granulocytes Colony-forming ability assay to evaluate the function of mouse hematopoietic progenitor cells.
Results: The ratio of HSC and LT-HSC in KKAy mice before irradiation was lower than that in C57 mice. After 4 Gy total body irradiation, the peripheral blood WBC, RBC, PLT, HGB and LYM% of KKAy mice decreased by 68.42% and 12.17%, respectively. %, 8.78%, 30.12%, 70.84%; the proportions of HPC, HSC and LT-HSC in the bone marrow decreased by 34.02%, 29.49% and 35.74%, respectively; the proportions of B cells and T cells in the spleen decreased by 57.85% and 58.81%, respectively. ;The proportion of CD4CD8 double-positive cells in the thymus decreased by 51.70%. The decrease in bone marrow HSC, LT-HSC, peripheral blood RBC and HGB of KKAy mice was significantly lower than that of C57 mice.
Conclusion: 4Gy whole body irradiation damages the hematopoietic immune system function of KKAy mice, and KKAy mice may show stronger tolerance to ionizing radiation than C57 mice.
