【Animal Modeling】-Quality Control of Chromosomal Specimen Preparation in Hamster In Vitro Chromosomal Aberration Test

  Objective: To investigate the influencing factors and quality control of the chromosome preparation process in the in vitro chromosome aberration test, and to summarize and analyze the successful preparation methods and key points of in vitro chromosome specimens.

  METHODS: Chinese hamster lung cells (CHL) were selected for cell culture and chromosome preparation. Mitomycin and cyclophosphamide were selected as positive mutagen. After routine hypotonicity, fixation, and dripping, the films were examined by microscopy.

  Results: In CHL chromosome aberration test, mitomycin and cyclophosphamide treatment significantly increased the chromosome aberration rate, all > 20%; while the negative control group, under the two test conditions with and without the metabolic activation system, the chromosome structure aberration rate was both Less than 5%. The prepared chromosomes are well dispersed, moderate in length and high in success rate.

  Conclusion: There are many factors that affect the preparation of chromosome specimens for in vitro chromosomal aberration test, and each step is very critical. Only by strictly following the operating procedures, mastering the principle of each step, and operating carefully, patiently, and scientifically, can a good specimen be prepared.
