【Animal modeling】-Inhibitory effect of atrial natriuretic peptide on the growth of transplanted melanoma in knockout mice

  Objective: To investigate the effect of atrial natriuretic peptide on the growth of melanoma.

  Methods: The melanoma B16F10 cells cultured in vitro were inoculated subcutaneously in ANP knockout mice and C57BL/6J mice, respectively, to establish a melanoma xenograft model. On the 12th day after tumor inoculation, the mice were sacrificed by neck dislocation, and the transplanted tumors were isolated and weighed. After paraffin-embedding the tumor tissue, the tumor cell proliferation and microvessel density were detected by immunohistochemistry.

  Results: The tumor volume of ANP knockout mice was significantly smaller than that of C57BL/6J mice, and the tumor weight was also significantly lighter. The number of cell proliferation and microvessel density in tumor tissues of ANP knockout mice were also significantly lower than those of C57BL/6J mice.

  Conclusion: ANP deletion may significantly inhibit the growth of melanoma by inhibiting tumor cell proliferation and microvascular formation.
