Objective: To compare the effects of different local intervention temperatures on endoplasmic reticulum stress (ERS) and cell apoptosis in rats, and to provide experimental basis for clinical prevention and treatment of pressure ulcers.
Methods: The pressure ulcer model was established by ischemia-reperfusion method, and 40 SPF adult male SD rats were randomly divided into 4 groups: sham operation group (sham group), model group, heat intervention group and cold intervention group. The muscle tissue of the rats in each group was cut on ice, and HE staining was used to observe the pathological changes of skeletal muscle; Western blot was used to detect the expression levels of endoplasmic reticulum stress-related proteins GRP78, casPase-12 and CHOP; immunofluorescence was used to observe the casPase- 12 and CHOP expression; TUNEL staining to observe cell apoptosis?
Results: Compared with the model group, the injury degree of skeletal muscle cells in the heat intervention group was aggravated, and the number of apoptotic cells increased. The immunofluorescence results showed that the positive expressions of casPase-12 and CHOP were increased. Compared with the model group, the expression levels were increased (P < 0.05); while the injury degree of skeletal muscle cells in the cold intervention group was reduced, and the apoptotic cells were reduced. Compared with the model group, the expressions of 12 and CHOP were decreased (P < 0.05)?
Conclusion: Local cold intervention can alleviate pressure ulcer injury in rats by inhibiting the apoptosis pathway mediated by endoplasmic reticulum stress; local thermal intervention aggravates ERS and promotes the occurrence of apoptosis, aggravating pressure ulcer injury in rats?
